The way he did it looks rather simple and different from the manual on here.

He also doesn't seem to have a regulator..

I also have the same tank and the same setup.

My question is:
Would it work in theory? With no regulator?
And how much helium is enough?

Because I only have one of them, [could by a second one].

The theory says that breathing oxygen at concentrations below 6% (at normal atmospheric pressure) causes rapid loss of consciousness and death in a few minutes. As long as you don't allow a presence of too much oxygen inside the bag, it doesn't matter how exactly you've achieved this. Technically, it's possible to create a good oxygen-depleted environment without constant gas flow. There is a widespread myth that CO2 must be constantly removed from the bag, but I've never seen any decent evidence in support of it. The guy on the picture used a large bag with a large volume of an asphyxiant gas inside, so somewhat significant CO2 buildup would take minutes there. On the other hand, breathing a gas mixture with only 5% O2 may produce unconsciousness and the surgical stage of anesthesia in 75 - 95 seconds. If you fall in coma before hypercapnic response is developed, CO2 probably should not be an issue.
